目的 建立测定盐酸替罗非班光学异构体的高效液相色谱方法。方法 采用ULTRON ES-OVM(4.6 mm×150 mm,5 μm)手性柱,以8%(m/m)醋酸铵缓冲液(pH 5.0)-40%(V∶V)的甲醇乙腈溶液(95∶5)为流动相,流速0.8 mL·min-1,柱温30 ℃,检测波长227 nm。结果 盐酸替罗非班光学异构体在0.4~10 μg·mL-1内均呈良好的线性关系,检测限为2 ng,定量限为6 ng,平均加样回收率为99.8%,与其D-对映体的分离度为2.1。结论 本方法简便、准确、灵敏,专属性强,适用于盐酸替罗非班光学异构体的测定。
Abstract
OBJECTIVE To develop a chiral HPLC method for the determination of enantiomers of tirofiban hydrochloride. METHODS The chromatographic conditions were as follows:ULTRON ES-OVM chiral column (4.6 mm×150 mm,5 μm) was adopted, 8%(m/m) ammonium acetate buffer (pH 5.0)-40%(V∶V) methanol in acetonitrile solution (95∶5) was used as the mobile phase eluted at 0.8 mL·min-1,the column temperature was 30 ℃, and the detection wavelength was set at 227 nm.RESULTS The calibration curves were linear over 0.4-10 μg·mL-1. The lowest limit of detection was 2 ng and the limit of quantitation was 6 ng. The average recovery was 99.8%. The enantiomers of tirofiban were separated with a resolution of 2.1. CONCLUSION This method is convenient, accurate, sensitive and specific for determination of enantiomers of tirofiban.
